Are some design variations that has been discussed by people and I think that we could use these variations to our advantage in Mars Direct and other developments including increasing personnel volumes in orbit and other large satellites and probes etc.

Look at the information provided on these SpaceRef News Pages:

Re: CEV Cargo and Crew Design Variations

No! No multitude of variations! Certainly no more than four versions total:
-HLV with upper stage
-HLV without upper stage
-TheStick five segment
-TheStick four segment

Re: CEV Cargo and Crew Design Variations

With ATK being the only game in town for the first stage they are looking to add to what they can do for the vision.

Opportunities for additional scope on space exploration missions include crew escape designs, lunar-lander systems, thermal and radiation shielding, and advanced lightweight materials

Hopefully ATK will start pursuing liquid engines in the future as well.

Re: CEV Cargo and Crew Design Variations

A few of them are interested in hypersonics--and their knowledge of chemical mixes allowed them to have a solar sail mock-up. I'd say ATK has come along quite nicely. They can drop the hypersonic nonsense though.

Focus on big rockets, and forget everything else for now.

Re: CEV Cargo and Crew Design Variations

With Nasa wanting to bow out from the ISS in more ways than one it is looking forward to the day when Entrepreneurs could take over job of sending cargo and crew into orbit
NASA seeks help from private rocketeers

Companies have until Feb. 10 to submit proposals to NASA for its transport services. The agency expects to award one or more contracts in May. NASA has allotted $500 million to pay for the initial phases of the program through 2010.

Here is a listing of companies that seem to want to do this for Nasa it is an excel file.
